In Vitro Propagation of Sansevieria laurentii from Leaf Explants


Abstract
In order to determine the possibility of micropropagation from leaf explants of Sansevieria laurentii by in vitro culture and to assess the mutation of periclinal chimera layer of the plantlets derived from explants cultured, explants taken from various part of leaf were cultured in vitro on Murashige-Skoog¢¥s medium supplemented with different concentrations of NAA or kinetin alone. And the results are summarized as follows.
Analysis of leaf explants indicated that N, P, K, Ca and Mg content in the yellow part of leaf was higher than that of green or light green part of leaf. The macronutrients content was similar between light green and green part of leaf.
Shoot differentiation from explants has tendency to delayed in all treatments. The best desired results for shoot differentiation obtained on the medium added 1.5§·/§¤ NAA. Shoot formed from yellow part of leaf tinged with green color. Shoot differentiation was completely inhibited on the medium supplemented with kinetin in any case.
Callus formation from explants cultured on the medium added 0.5§·/§¤ kinetin of 1.5§·/§¤ NAA, which was optimum concentrations for root formation, was favorable except yellow part of leaf.
There was no differences between light green and green part of leaf in callus, shoot, and root formation.
